Product details:
3' to 5' Exonuclease Activity Assay Kit (Fluorometric) (ab273269) provides a quick and easy method for monitoring 3' exonuclease activities in a wide variety of samples. In this assay, exonuclease digests the provided DNA probe, producing a strong fluorescent signal (Ex/Em = 304/369nm). The kit is simple, sensitive, high-throughput adaptable and can detect as low as 0.2 μU of exonuclease activity.
This product is manufactured by BioVision, an Abcam company and was previously called K175 3' to 5' Exonuclease Activity Assay (Fluorometric). K175-100 is the same size as the 100 test size of ab273269.
The Safety Datasheet for this product has been updated for certain countries. Please check the current version in the Support and downloads section.

The 3' to 5' exonuclease also known as exonuclease III is an important enzyme that removes nucleotides from the ends of a DNA strand in the 3' to 5' direction. Its role in DNA repair and proofreading contributes to maintaining genomic stability. Exonuclease III is expressed primarily in the cell nucleus and has a molecular mass of about 31 kDa. The enzyme's activity is essential for DNA replication and repair processes and it displays strong exonuclease activity.
Biological function summary
Exonuclease removes mismatched bases during DNA proofreading therefore ensuring the accuracy of DNA replication. It acts as part of the DNA polymerase complex where it helps maintain the fidelity of DNA synthesis. The enzyme also participates in base excision repair (BER) pathways a critical mechanism for correcting DNA single-strand breaks. By participating in these processes exonuclease plays a part in preserving the genome integrity of the cell.
Pathways
Exonuclease 3' to 5' activity is important in the mismatch repair (MMR) and nucleotide excision repair (NER) pathways. In these pathways the exonuclease partners with DNA polymerase and other nucleases like endonucleases ensuring the replacement of incorrect or damaged nucleotides. It works synergistically with proteins such as DNA ligase which seals the nicks in the DNA phosphodiester backbone. Through these collaborative interactions exonuclease supports the dynamic regulation of DNA integrity.
Exonuclease's function intertwines with oncogenesis and neurodegenerative diseases. Its deficiency or malfunction can lead to conditions like cancer due to the accumulation of mutations from failed DNA repair mechanisms. The 3' to 5' exonuclease activity insufficiency is linked with proteins such as proliferating cell nuclear antigen (PCNA) which are vital in DNA replication and repair processes. In neurodegenerative diseases compromised exonuclease activity leads to an increase in DNA damage contributing to disease progression.
