Phospho-RSK2 (S386) ELISA Kit (ab279916) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated RSK2 (S386) protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an
in vitro
enzyme-linked immunosorbent assay for the measurement of human phospho-RSK2. An anti-pan RSK2 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and RSK2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-RSK2 (S386) antibody is used to detect phosphorylated RSK2. After washing away unbound antibody, HRP conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RSK2 (S386) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

RSK2 also known as ribosomal S6 kinase 2 is a serine/threonine-protein kinase with a molecular mass of approximately 85 kDa. This protein is expressed in various tissues with significant levels in the brain lungs and muscles. Mechanically RSK2 phosphorylates a variety of substrates involved in cell cycle regulation transcription and growth linking extracellular signals to intracellular responses. It contains two non-identical kinase domains which interact and modulate its activity leading to changes in cellular dynamics.
Biological function summary
RSK2 influences cell proliferation growth and survival by acting as a bridge in signaling cascades downstream of the extracellular signal-regulated kinase (ERK) pathway. This protein is not part of a larger complex but it interacts with other proteins such as ERK1/2 and substrates like ribosomal protein S6.
