) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Monkey TIM-3 protein in monkey srum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Monkey TIM-3 with 7.77 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. T-Cell Membrane Protein 3 (TIM-3), also known as Hepatitis A Virus Cellular Receptor 2 is type 1 transmembrane protein encoded by the
HAVCR2
gene. TIM-3 contains a 181- amino acid extracellular portion that includes a Ig-like V-type domain, a 21- amino acid- transmembrane section, and a 78- amino acid cytoplasmic section. TIM-3 is a cell surface receptor expressed in Th1 lymphocytes, regulatory T-cells, innate immune cells including dendritic cells, NK cells, and monocytes. Binding to Galectin-9 induces intracellular calcium influx which triggers apoptosis. Other ligands are phosphatidyl serine, High Mobility Group Protein 1 (HMGB1), and Carcinoembryonic Antigen Related Cell Adhesion Molecule 1 (CEACAM1). TIM-3 can be cleaved to a soluble form from the cell surface by ADAM10 or ADAM17 mediated lysis. The soluble form of TIM-3 has been found to be elevated in the plasma of patients with HIV/AIDS, Hepatitis, and graft-versus-host Disease.

TIM-3 also known as T cell immunoglobulin and mucin-domain containing-3 or HAVCR2 is a protein involved in immune regulation. It possesses an approximate mass of 35 kDa. TIM-3 is expressed on various immune cells including T cells NK cells and dendritic cells especially after activation. The expression level often changes in response to inflammatory conditions suggesting its role in modulating immune responses.
Biological function summary
TIM-3 functions as a checkpoint inhibitor impacting immune cell activity. It is not part of a larger physical complex but it modulates immune responses by interacting with its ligands such as Galectin-9 phosphatidylserine and CEACAM1. TIM-3 involvement in downregulating Th1 cell responses shows its necessary role in maintaining immune homeostasis.
