SARS-CoV-2 nsp8 also known as non-structural protein 8 forms part of the viral replicase complex of the severe acute respiratory syndrome coronavirus 2. Nsp8 has a mass of approximately 22 kDa and plays an important role in the RNA synthesis process of the virus. It is expressed inside host cells once the virus enters the host organism. The protein functions as a part of the RNA polymerase complex working alongside other non-structural proteins to enable the virus to replicate its genetic material effectively.
Biological function summary
Nsp8 cooperates with non-structural protein 12 (nsp12) to form the RNA-dependent RNA polymerase (RdRp) complex. This complex is essential for the replication and transcription of the viral genome. Nsp8 nsp12 and nsp7 come together as a multi-protein complex enhancing the activity of the RdRp. Nsp8 has been shown to stabilize the structure and function of the replication machinery providing the necessary support for the synthesis of the viral RNA strand.
Pathways
Nsp8 plays an important role in the viral replication and transcription pathways. The polymerase activity driven by the complex of nsp8 nsp12 and nsp7 operates through the replication-transcription complex important for efficient viral propagation within the host. This process not only involves the formation of viral RNA genomes but also subgenomic RNAs which are required for viral protein translation. The interaction between these non-structural proteins ensures that the virus maintains a high level of infectious presence in its host.
Nsp8 holds a strong connection to COVID-19 the disease caused by SARS-CoV-2 infection. The interplay between nsp8 and other non-structural proteins such as nsp12 contributes directly to the ability of the virus to replicate and cause widespread infection. Inhibition of nsp8 function could provide a therapeutic target for treating COVID-19 as disruption of the RdRp complex could hinder the viral replication process. Given the pivotal role of this protein in the virus’s lifecycle targeting nsp8 could offer promising avenues for antiviral drug development.
