Product details:
The Human CPLX1 ELISA Kit (ab283984) is designed for detection of complexin-1 (CPLX1) in human cell lysate and tissue samples. This assay employs a quantitative sandwich enzyme immunoassay technique that measures human complexin-1 in approximately 5 hours.
A polyclonal antibody specific for human complexin-1 has been pre-coated onto a 96-well microplate with removable strips. CPLX1 in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for human complexin-1, which is recognized by a streptavidin-peroxidase (SP) conjugate. All unbound material is washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.

The target CPLX1 also known as Complexin-1 functions in the mechanical control of presynaptic vesicle release. It weighs approximately 16 kDa and finds expression primarily in the central nervous system. CPLX1 interacts with the SNARE complex playing a role in neurotransmitter release. This protein ensures precise regulation at synaptic junctions which are important for efficient neuronal communication.
Biological function summary
Complexin-1 acts as an essential regulator of synaptic exocytosis. It stabilizes the SNARE complex inhibiting premature synaptic vesicle fusion while also preparing the vesicles for rapid release upon calcium influx. In synaptic terminals Complexin-1 ensures quick and precise neurotransmitter release therefore supporting synaptic plasticity and signal transduction essential for learning and memory. Complexin-1 forms complexes with proteins like synaptobrevin and syntaxin participating in the excitatory and inhibitory synaptic transmission.
Pathways
Complexin-1 plays a significant role in the synaptic vesicle cycle and neurotransmitter secretion pathways. It interacts with proteins in the SNARE complex notably SNAP-25 and synaptotagmin facilitating neurotransmitter release upon stimulus. Its function in these pathways supports the maintenance of synaptic efficacy and neuronal circuitry critical for normal nervous system operation and responses to environmental stimuli.
Abnormalities in Complexin-1 expression or function link to neurological conditions like epilepsy and schizophrenia. Both of these disorders involve disrupted synaptic transmission and altered neurotransmitter release. Additionally Complexin-1 has connections with the protein alpha-synuclein implicated in neurodegenerative diseases such as Parkinson's disease. These associations highlight the importance of Complexin-1 in maintaining synaptic function and its potential as a therapeutic target.
