UNC5C also known as Unc-5 Netrin Receptor C serves as a receptor for netrin a protein that guides axonal growth. It is a member of the UNC5 family and plays an important role in cell migration and positioning. The molecular mass of UNC5C is approximately 103 kDa. It is mainly expressed in the nervous system specifically in the developing brain and spinal cord but can also be found in other tissues to a lesser degree. Its expression pattern suggests a role diverse from just the nervous system.
Biological function summary
UNC5C functions significantly in neuronal guidance and apoptosis regulation. It forms part of a receptor complex together with DCC (Deleted in Colorectal Carcinoma) when binding with netrin-1 another important protein in axon guidance. This interaction influences neuronal cells’ behavior by repelling or attracting axons depending on the context. Beyond its interaction with netrin-1 UNC5C also engages in signaling pathways promoting or inhibiting cell death adding to its versatile functions in cellular processes.
Pathways
UNC5C participates in the axon guidance and apoptotic processes. It plays an important role in the netrin signaling pathway by interacting with DCC and netrin-1 helping to steer neuron growth cones during development. Additionally it has involvement with the apoptosis pathway where UNC5C interactions can lead to programmed cell death when certain conditions are met such as absence of netrin-1. The protein's function is intricately linked with others like DCC and the netrin family which means it influences and is influenced by key neurodevelopmental pathways.
Mutations or dysregulation of UNC5C have been associated with neurodegenerative conditions and cancer. Altered UNC5C signaling can contribute to the progression of Alzheimer's disease potentially through the disruption of normal axonal pathfinding or neuronal death mechanisms. In cancer particularly colorectal cancer UNC5C acts as a dependence receptor where its malfunction can trigger inappropriate cell survival contributing to tumorigenesis. UNC5C’s interaction with DCC highlights its role in colorectal cancer emphasizing how its impairment can promote the escape from normal apoptotic signals.
