PIP5K1 alpha also known as Phosphatidylinositol-4-phosphate 5-kinase type 1 alpha (PIP5K1A) is an enzyme that catalyzes the phosphorylation of phosphatidylinositol-4-phosphate (PI4P) to generate phosphatidylinositol-45-bisphosphate (PI(45)P2). The protein has an approximate mass of 68 kDa and is highly expressed in a variety of human tissues including the brain heart and spleen. PIP5K1A plays a significant role in lipid signaling and membrane dynamics impacting a wide array of cellular processes such as cytoskeletal organization and vesicle trafficking.
Biological function summary
PIP5K1A influences the production of PI(45)P2 an important phosphoinositide involved in numerous cellular functions. PI(45)P2 acts as a substrate for the production of secondary messengers like inositol trisphosphate (IP3) and diacylglycerol (DAG) essential in signal transduction pathways. PIP5K1A is part of a larger protein complex interacting with cytoskeletal elements and regulators of endocytosis and exocytosis. This association highlights its importance in maintaining cellular compartmentalization and function.
Pathways
PIP5K1A plays a pivotal role in the phosphoinositide signaling pathway and the actin cytoskeleton regulation pathway. It works closely with proteins such as RAC1 a small GTPase involved in actin polymerization to facilitate changes in the cell structure and motility. Additionally PIP5K1A's product PI(45)P2 serves as a precursor for multiple signaling molecules integrating with pathways that mediate cellular responses to various extracellular stimuli.
PIP5K1A's dysregulation links to cancer and neurological disorders. In cancer alterations in PIP5K1A expression or activity can affect cell proliferation and survival interacting with proteins like AKT that drive oncogenic signaling. In the context of neurological disorders its role in synaptic function and plasticity directly connects to conditions like schizophrenia where PI(45)P2 imbalance may contribute to the disorder's pathophysiology. These associations highlight the essential impact of PIP5K1A in maintaining cellular homeostasis and its potential as a therapeutic target.
