The Nogo Receptor also known as Reticulon 4 receptor (RTN4R) is a protein that functions as an axon growth inhibitor. This receptor with an approximate molecular mass of 55 kDa binds to myelin-associated inhibitors to regulate neuronal growth and regeneration. It is part of the Nogo signaling pathway and is expressed mainly in the central nervous system. The receptor interacts with ligands like Nogo-A MAG and OMgp which play key roles in the restriction of axonal regeneration.
Biological function summary
Nogo Receptor plays a critical role in the regulation of neural plasticity and axonal growth. The receptor forms a complex with co-receptors such as p75 neurotrophin receptor (p75NTR) and LINGO-1 and this complex significantly influences signal transduction. Nogo Receptor-mediated signaling restricts the ability of neurons to regrow or extend after injury maintaining inhibitory influence on the nervous system regeneration.
Pathways
This receptor actively participates in neural inhibitory signaling and axon guidance pathways. It integrates signals from proteins like Nogo-A and glial inhibitors to regulate neuron morphology and network architecture. Understanding these pathways reveals Nogo Receptor's involvement in neuronal development and injury responses associating with signaling proteins like ROCK that mediate structural rearrangement of the cytoskeleton.
Nogo Receptor has relevance particularly in central nervous system injuries and multiple sclerosis (MS). Its high expression in the CNS aligns with limited neural repair and recovery in these conditions. Nogo Receptor signaling inhibits axonal regeneration contributing to the pathology of diseases like spinal cord injury and MS. In multiple sclerosis interactions between Nogo Receptor and MAG or OMgp correspond with the restricted repair and remyelination processes observed in demyelinating lesions.
