ESAM also known as Endothelial Cell-Selective Adhesion Molecule is part of the immunoglobulin superfamily and is typically found on the surface of endothelial cells. It has a mass of roughly 44 kDa. The ESAM protein is expressed predominantly in vascular and lymphatic endothelial cells but can also be found in platelets. ESAM is important for cell adhesion playing a role in maintaining cell junctions. Through interaction with other proteins ESAM contributes to the structural integrity of blood vessels.
Biological function summary
ESAM influences vascular stability and permeability. It participates in the formation of endothelial tight junctions being an important component of cell junction complexes. In endothelial cells ESAM enhances barrier function impacting the movement of molecules and cells between the bloodstream and surrounding tissues. Its expression is essential for the organization of other junctional proteins such as claudins and occludins indicating a synergistic role within cell junctions.
Pathways
The ESAM protein is involved in the regulation of vascular integrity and immune response pathways. Particularly it interacts with signaling components of the vascular endothelial growth factor (VEGF) pathway which is important in angiogenesis. Additionally ESAM modulates the activation of PI3K/Akt signaling promoting cell survival and proliferation. Through its associations ESAM works alongside other proteins such as VE-cadherin highlighting its integrative role in vascular function.
ESAM has shown a connection to cancer progression and inflammatory diseases. In tumors aberrant ESAM expression correlates with increased tumor angiogenesis and metastasis partly due to its influence on endothelial cell permeability and migration. ESAM also relates to atherosclerosis interacting with proteins like ICAM-1 which are involved in leukocyte adhesion and vascular inflammation. Understanding its expression and function in these conditions can help in developing targeted therapies.
