Product details:
Human FCRL3 (FcRH3) ELISA kit is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of human FCRL3 (FcRH3) in serum (human FCRL3 concentration is low in normal serum/plasma, and may not be detectable in this assay), plasma (collect plasma using EDTA and Citrate as an anticoagulant. Heparin is not recommended in this assay), and cell culture supernatants. This assay employs an antibody specific for human FCRL3 (FcRH3) coated on a 96-well plate.
This ELISA kit shows no cross-reactivity with the following cytokines tested: human ANGPTL7, CD36, CLEC9a, CL-P1, Dectin-2, DLL4, DSCAM, EDIL3, ENPP-7, Enteropeptidase, FCRLB, FGF-3, FLRT1, FLRT2, GBA3, GDF-11, Glycoprotein V, Granzyme A, IGSF4B, IL-28 R alpha, Kynureninase, LAMA4, LRRC4, LRRTM3, NG2, NQO-1, PCSK2, PILR-alpha, Plexin A4, POGLUT1, PRELP, Smad4, SOX15, SOX7, SOX9, Syntaxin 6, TROP2, TSLP R, UNC5H4.

FCRL3 also known as Fc receptor-like 3 is a protein encoded by the FCRL3 gene. It weighs approximately 75 kDa and plays an important role in immune regulation. FCRL3 is predominantly expressed on B cells but it is also found on T cells and natural killer cells. The protein is part of the family of Fc receptor homologs and features immunoreceptor tyrosine-based activation motifs (ITAMs) and immunoreceptor tyrosine-based inhibitory motifs (ITIMs). These motifs allow FCRL3 to function both as an activator and inhibitor of immune responses depending on the context.
Biological function summary
FCRL3 acts in regulating immune cell activation and differentiation. It influences the balance of auto-reactive and protective immune responses. FCRL3 does not form multimeric complexes but interacts with other immune receptors and signaling proteins. By modulating signaling pathways it impacts processes such as antibody production cellular proliferation and cytokine release. The protein contributes to maintaining immune homeostasis and preventing excessive inflammatory responses.
Pathways
FCRL3 engages in immune signaling cascades notably the B cell receptor (BCR) pathway and the T cell receptor (TCR) pathway. These pathways are central to adaptive immunity involving receptor signaling and subsequent cell activation. FCRL3 interacts with other proteins such as SYK and SHP-1 which mediate downstream signaling events in these pathways. This modulation is important for the fine-tuning of adaptive immune responses influencing immunity against pathogens and tolerance towards self-antigens.
FCRL3 has been associated with autoimmune conditions like rheumatoid arthritis and systemic lupus erythematosus. In these disorders FCRL3 expression influences pathogenic B cell activity and autoantibody production. The protein shares interactions with CD20 and BLIMP-1 both key regulators of B cell function which link alterations in FCRL3 activity with disease progression and severity. Understanding FCRL3's role in these conditions aids in developing targeted therapies for autoimmune diseases.
