Gm6A also known as N6-methyladenosine (m6A) methyltransferase is an essential component of the RNA modification machinery. It plays a critical role in post-transcriptional regulation. The protein mass of Gm6A is approximately 86 kDa. It is widely expressed in mammalian cells particularly prominent in brain and liver tissues. Gm6A facilitates the methylation of adenosine residues on RNA molecules influencing RNA stability splicing and export.
Biological function summary
Gm6A functions in the regulation of RNA metabolism. As an important player in epitranscriptomics it is part of the methyltransferase complex along with METTL3 and METTL14. This complex modifies messenger RNA to control gene expression and ensure proper cellular responses to various stimuli. The modification process governed by Gm6A is essential for maintaining RNA dynamic functions involving feedback loops where altered RNA impacts protein synthesis and cell signaling pathways.
Pathways
Multiple cellular processes rely on the functions of Gm6A methylation. It's involved in the mRNA processing pathway contributing to splicing and alternative polyadenylation. Gm6A interacts with proteins like YTHDF1 and YTHDF2 which mediate mRNA translation and degradation. Through these pathways Gm6A supports cell differentiation and stress responses embedding itself in the larger network of gene expression regulation.
Gm6A regulation has implications in cancer and neurological disorders. Altered m6A methylation patterns correlate with tumorigenesis in cancers including leukemia and glioblastoma. The aberrant methylation affects proteins like FTO which is an m6A demethylase potentially driving oncogenic pathways. In neurological contexts disruptions in m6A methylation play a role in neurodevelopmental disorders with implications for proteins such as RBM15 that are involved in cognitive functions and brain development.
