Desmoplakin is a critical component of the cell structure acting as a link between desmosomes and the intermediate filament network. It exists in two isoforms Desmoplakin I and Desmoplakin II with molecular masses of approximately 332 kDa and 260 kDa respectively. Desmoplakin is mainly expressed in tissues with high mechanical stress such as the heart and skin where strong cell adhesion is necessary. Alternate names like Desmoplakine may also be used in some scientific literature.

It serves as part of the desmosomal complex where it anchors intermediate filaments to the desmosomal plaque ensuring the mechanical stability required by tissues under mechanical stress. Mutations or dysfunctional interactions of desmoplakin can lead to conditions like arrhythmogenic right ventricular cardiomyopathy (ARVC) and skin disorders such as epidermolysis bullosa simplex. In ARVC desmoplakin-related defects may disrupt interactions with other desmosomal proteins like plakoglobin impacting heart muscle integrity and function. Similarly in epidermolysis bullosa simplex altered desmoplakin function compromises skin cell adhesion leading to increased susceptibility to injury.
