FIG4 also known as SAC3 or Fleckstein1 is a phosphoinositide phosphatase enzyme with a mass of approximately 104 kDa. It is expressed in various tissues including the brain and muscle cells. Mechanically FIG4 functions to dephosphorylate phosphoinositides important molecules involved in membrane dynamics and intracellular signaling. By regulating the levels of these phosphoinositides FIG4 modulates cellular processes such as trafficking and autophagy contributing to proper cellular homeostasis.
Biological function summary
FIG4 engages in maintaining lysosomal function and endocytic membrane trafficking operating within a specific protein complex with VAC14 and PIKFYVE. This complex plays a role in the synthesis and turnover of phosphoinositide 35-bisphosphate (PI(35)P2) a lipid that modulates lysosome membrane stability and endosomal sorting. These activities are vital for cellular nutrient recycling and responding to cellular stress impacting cellular remodeling and health.
Pathways
FIG4 is involved in the phosphoinositide signaling pathway impacting the turnover of PI(35)P2. It connects with the autophagy pathway influencing lysosome biogenesis and function. Through these pathways FIG4 interacts with proteins like PIKFYVE and VAC14 forming a regulatory framework that ensures proper lysosomal degradation and membrane-related signaling required for cellular adaptation and response to environmental changes.
FIG4 mutations or dysregulation link to conditions such as Charcot-Marie-Tooth disease type 4J and amyotrophic lateral sclerosis (ALS). In these disorders altered levels of PI(35)P2 lead to abnormal lipid metabolism and cellular transport affecting neuron function and survival. The interactions with proteins like PIKFYVE and other phosphatases emphasize FIG4's role in maintaining neuronal integrity and its potential involvement in neurodegenerative disease progression.
