Endothelial cell-specific molecule 1 (ESM1) also known as endocan is a proteoglycan with a mass of approximately 50 kDa. It is primarily expressed in endothelial cells of blood vessels particularly in the lungs and the kidneys. ESM1 consists of a core protein covalently attached to dermatan sulfate chains. Its expression often increases under inflammatory conditions which highlights its role in vascular biology.
Biological function summary
ESM1 acts in the regulation of cell adhesion and migration processes. It interacts with leukocytes and participates in modulating inflammatory responses. ESM1 does not form part of a multi-protein complex but its activity can influence the behavior of other cell surface molecules and receptors. This function positions ESM1 as a critical player in the dynamics of vascular endothelium.
Pathways
ESM1 modulates inflammation and angiogenesis by participating in pathways involving vascular endothelial growth factor (VEGF). It interacts with proteins such as VEGF receptor-2 impacting endothelial permeability and new blood vessel formation. These interactions situate ESM1 within the signaling networks that regulate endothelial function and vascular homeostasis.
Research associates ESM1 with conditions such as sepsis and cancer. High levels of this molecule correlate with poor prognosis in these diseases linking it functionally to VEGF and other factors involved in abnormal endothelial cell activity. Understanding ESM1's role provides insights into mechanisms of disease progression and potential therapeutic targets.
