MLN64 also known as StAR-related lipid transfer domain protein 3 (STARD3) plays an important role in intracellular cholesterol transport. It has a molecular weight of approximately 50 kDa. MLN64 is mainly expressed in the adrenal gland placenta and various cancerous tissues. As a transmembrane protein located in the endoplasmic reticulum and late endosomes it facilitates lipid transport between membranes by binding cholesterol through its START domain.
Biological function summary
MLN64 influences cholesterol homeostasis by transporting cholesterol from late endosomes to the mitochondria. It acts outside the context of a known protein complex yet exerts significant control over cholesterol regulation within cells. By controlling cholesterol distribution MLN64 supports membrane synthesis steroid production and cellular signaling processes. Its function is integral to maintaining lipid balance within different cell types.
Pathways
MLN64 is essential for lipid metabolism and steroid biosynthesis. It directly links with the cholesterol trafficking pathway where it interacts with proteins such as Niemann-Pick C1 (NPC1). This interplay facilitates cholesterol efflux from late endosomes a critical step in cellular lipid regulation. Additionally MLN64's activity affects the steroidogenic pathway aiding in the conversion of cholesterol into steroidal hormones through cooperation with hormone-sensitive lipase.
MLN64 has connections to conditions such as Niemann-Pick disease and steroidal imbalances. Dysfunctional cholesterol transport often involving NPC1 can manifest in Niemann-Pick disease illustrating MLN64's important involvement. Also altered MLN64 activity affects steroid hormone levels contributing to disorders related to abnormal steroid synthesis. Understanding MLN64's multifaceted role in these conditions may offer insights for therapeutic interventions.
