Hepatitis C Virus genotype 1a NS5 also called non-structural protein 5 plays an important role in viral replication. The NS5 protein encompasses two important domains: NS5A and NS5B with a molecular weight of roughly 100 kDa. It is expressed in infected host cells and its functional activities are primarily associated with the hepatitis C replication complex. This complex resides on the endoplasmic reticulum membranes within the host cell cytoplasm which facilitates the efficient replication of the viral RNA.

The NS5 protein is indispensable for the formation and stabilization of the replication complex.
