The insulin receptor substrate 4 (IRS4) is a protein that plays a significant role in transmitting signals from insulin and insulin-like growth factor receptors to downstream pathways. It has a molecular mass of approximately 130 kDa. IRS4 is mainly expressed in the hypothalamus pituitary gland and several other organs such as liver and adipose tissues. IRS4 interacts with various signaling intermediates through its phosphotyrosine-binding domains therefore acting as a docking protein that facilitates signaling cascades.
Biological function summary
The role of IRS4 extends beyond signal transduction. It participates in the regulation of growth survival and metabolism. Although not part of a permanent protein complex IRS4 temporarily binds to receptors and adaptor proteins during signal transduction processes. The interactions allow IRS4 to integrate signals from different receptors to produce coordinated cellular responses particularly in energy homeostasis and glucose metabolism.
Pathways
IRS4 serves important functions in the insulin signaling and growth hormone signaling pathways. Within the insulin signaling pathway IRS4 acts as a central node linking insulin receptors to the activation of phosphoinositide 3-kinase (PI3K) which subsequently leads to activation of protein kinase B (AKT) and mitogen-activated protein kinase (MAPK) pathways. Related proteins in these cascades include other insulin receptor substrates such as IRS1 and IRS2 which share overlapping roles but may exhibit tissue-specific functions.
IRS4 exhibits associations with metabolic conditions such as diabetes and obesity. Alterations in IRS4 expression or function can disrupt normal insulin signaling contributing to insulin resistance and metabolic dysregulation. IRS4 also shows connectiveness with proteins like PI3K and AKT which are frequently implicated in these metabolic disorders. Dissection of IRS4's role could potentially offer insights for therapeutic strategies targeting these diseases.
