Neuritin sometimes called cpg15 is a glycosylphosphatidylinositol (GPI)-anchored protein with a molecular mass of about 15 kDa. It is mainly expressed in the central nervous system particularly in areas involved with learning and memory including the hippocampus and the cerebral cortex. Neuritin plays a role in the development and plasticity of neurons assisting in processes like neurite outgrowth which supports neural connectivity and regeneration.
Biological function summary
Neuritin influences neuronal development by promoting neurite extension and synaptic remodeling. It has not been identified as part of a larger complex but interacts with cell surface molecules to exert its effects. Neuritin supports synapse formation and stabilization which are important for effective synaptic transmission and plasticity in neurons. This function gives neuritin a significant role in the regulation of neural circuits that underlie learning and cognitive functions.
Pathways
Neuritin plays roles in the neural growth and plasticity pathways. It has been associated with the MAPK/ERK signaling pathway influencing synaptic function and structure. Neuritin also interacts with small GTPases which are involved in axonal growth and dendritic branching. These interactions with other proteins help to mediate its effects on neuron structure and function contributing to overall neural network adaptability.
Neuritin has links to neurological conditions such as Alzheimer's disease and major depressive disorder. Its expression levels change in response to these diseases suggesting a protective or compensatory role. Neuritin interacts with amyloid precursor protein (APP) in Alzheimer's disease possibly influencing amyloid-beta production and plaque formation. In major depressive disorder interactions with brain-derived neurotrophic factor (BDNF) may affect mood regulation and synaptic health reflecting neuritin's potential impact on disease progression and symptomatology.
