The Lethal Factor also known as Anthrax Lethal Factor is an important component of the anthrax toxin produced by Bacillus anthracis. It has a molecular mass of approximately 90 kDa. This protein operates as a zinc-dependent metalloprotease and cleaves specific mitogen-activated protein kinase kinases (MAPKKs). The Lethal Factor along with Protective Antigen and Edema Factor form the tripartite Anthrax Toxin complex. While the Protective Antigen enables entry into host cells the Lethal Factor executes the proteolytic function. It is predominately expressed in conditions where Bacillus anthracis infects mammalian hosts enabling its toxic potential.
Biological function summary
Lethal Factor interferes with intracellular cell signaling pathways important for immune response. By specifically targeting MAPKKs it disrupts the regulation of critical cellular processes including cell growth differentiation and survival. The complex formed by Lethal Factor and Protective Antigen allows the toxin to enter the host cell and exert its deleterious effects. The enzymatic activity of Lethal Factor depends on its ability to enter cells where it performs in conjunction with other proteins like the Edema Factor.

Several cellular processes become affected by the Lethal Factor's activity. This protein disrupts the MAPK signaling pathway which is essential for cellular responses such as proliferation differentiation and stress response. Also it combines with Edema Factor to contribute to pathophysiology by amplifying cell dysregulation. By interacting with MAPKK enzymes the activity of downstream kinases such as ERK1/2 and JNK get inhibited which in turn affects cytokine production and cell survival mechanisms.
Lethal Factor is closely associated with Anthrax disease a zoonotic disease that primarily affects herbivorous animals but can infect humans. Lethal Factor contributes to the severity of the disease by impeding immune system function and disrupting cellular homeostasis. The protein interacts with Protective Antigen to form lethal toxin an important player in the pathogenesis of the anthrax disease. Through disruption of signaling pathways and immune modulation these two protein factors foster the development of systemic shock and mortality.
