ARNT2 also known as Aryl Hydrocarbon Receptor Nuclear Translocator 2 plays a role in transcription regulation by partnering with other bHLH-PAS proteins. Weighing approximately 88 kDa ARNT2 is mainly found in the brain and kidneys. This protein forms a heterodimer with other proteins such as HIF-1α which enables it to regulate gene expression in response to environmental signals.
Biological function summary
ARNT2 functions as a transcriptional regulator within cellular systems. It is a part of the hypoxia-inducible factor (HIF) complex which is involved in responding to low oxygen levels in cells. ARNT2 contributes to the activation of genes that help cells adapt to hypoxic conditions and it plays a role in neuronal development and function. The involvement of ARNT2 in these processes is critical for normal physiological functions.
Pathways
ARNT2 links closely with the hypoxia signaling pathway contributing to oxygen homeostasis. It interacts with proteins like HIF-1α to activate transcription of genes involved in angiogenesis metabolism and erythropoiesis. Another significant pathway involving ARNT2 is related to circadian rhythm regulation where it partners with CLOCK and BMAL1 proteins influencing daily biological cycles.
ARNT2 has been associated with conditions like neurodevelopmental disorders and certain types of cancer. Mutations or dysregulation of ARNT2 may connect to neurodevelopmental issues likely through its interactions with proteins such as HIF-1α impacting brain function. In cancer ARNT2's involvement in hypoxia pathways can affect tumor growth and survival linking it to potential therapeutic targets.
