DDX46 also known as DEAD-box helicase 46 is a protein involved in RNA processing and is part of the DEAD-box protein family. It has a molecular mass of approximately 103 kDa. Mechanically DDX46 functions as an ATP-dependent RNA helicase unwinding RNA secondary structures. This action is essential during RNA splicing events. DDX46 is expressed in various tissues including the liver testes and brain pointing to its importance in diverse cellular functions.
Biological function summary
DEAD-box helicases like DDX46 play a role in the biogenesis of ribosomal RNA and pre-mRNA splicing. DDX46 specifically contributes to the assembly of the U4/U6.U5 tri-snRNP complex a major component of the spliceosome. The spliceosome is a large and dynamic molecular complex important for the splicing of precursor messenger RNA transforming it into mature messenger RNA. DDX46's helicase activity facilitates RNA-RNA base pairing changes necessary for spliceosome assembly.

DDX46 acts within the RNA splicing pathway coordinating with other spliceosomal proteins such as PRPF31 and SNRNP200. These proteins work together to ensure accuracy and efficiency in the removal of introns from pre-messenger RNA.
