JPH3 also known as junctophilin-3 is a protein that plays a critical role in forming junctional membrane complexes between plasma membrane and endoplasmic or sarcoplasmic reticulum. This protein weighs approximately 82 kDa and helps in maintaining cellular calcium homeostasis. The expression of junctophilin-3 is mainly in the brain particularly within the neuronal tissues indicating its significant role in neural functions.
Biological function summary
Junctophilin-3 contributes to the structural foundation within cells by bridging intracellular membranes with the plasma membrane which is vital for maintaining the proper internal cell architecture. It is a part of a membranous network responsible for calcium ion signaling and plays a role in synaptic function influencing neuronal communication. This function supports the regulation of cellular processes tied to neural activity and development.
Pathways
Junctophilin-3 interacts with key biological pathways such as calcium signaling and neuronal signaling pathways. It connects with these pathways to ensure proper calcium influx necessary for neurotransmitter release and neuronal firing. JPH3 works alongside other proteins like junctophilin-1 in maintaining these pathways highlighting its significance in synaptic transmission and neural excitability.
Alterations in junctophilin-3 expression or function have been associated with neurodegenerative disorders such as Huntington's disease. Research indicates that JPH3 contains a CAG repeat region which when abnormally expanded leads to neuronal dysfunction and degeneration. Furthermore JPH3 interacts with proteins involved in Huntington's disease affecting cellular processes and contributing to the disease pathology.
