NUCKS1 also known as Nuclear Casein Kinase and Cyclin-dependent Kinase Substrate 1 is a chromatin-associated protein with a mass of approximately 27-31 kDa. It is ubiquitously expressed in human tissues but shows higher expression in certain cell types such as neurons and hepatocytes. NUCKS1 localizes to the nucleus and binds to chromatin influencing transcriptional processes.
Biological function summary
NUCKS1 acts as a transcriptional regulator in chromatin organization. It is not part of a complex but interacts with other chromatin modifiers to modulate gene expression. This protein affects cell cycle progression and DNA repair mechanisms where its influence is important for maintaining normal cellular functions.
Pathways
NUCKS1 is involved in the DNA damage response and cell cycle control pathways. It participates in the ATM/ATR pathway interacting with proteins such as BRCA1 to facilitate DNA repair. NUCKS1 also influences the transcriptional regulation pathways that alter the cell cycle dynamics whose proper functioning helps prevent unregulated cell growth.
NUCKS1 has links to cancer and neurodegenerative diseases. In cancer alterations in NUCKS1 expression may affect its role in cell cycle regulation with potential participation by the tumor suppressor protein p53 contributing to uncontrolled cell proliferation. In neurodegenerative diseases abnormal NUCKS1 expression can disrupt neuronal function potentially interacting with proteins like tau that are implicated in conditions such as Alzheimer's disease.
