DNA polymerase kappa also known as POLK or kappa polymerase is a specialized enzyme involved in DNA replication and repair. This protein weighs approximately 93 kDa. POLK is expressed across various human tissues with notable activity in testis and placenta. Mechanically DNA polymerase kappa participates in translesion synthesis a process that permits DNA replication to proceed past damaged or abnormal bases allowing cells to overcome blockages that conventional DNA polymerases cannot bypass.
Biological function summary
DNA polymerase kappa plays a significant role in error-prone repair mechanisms contributing to DNA damage tolerance. It is not known to associate directly with a larger complex but works in concert with other translesion synthesis polymerases such as DNA polymerase eta. Through its action POLK introduces nucleotides opposite DNA lesions maintaining replication fork progression and cell viability under genotoxic stress conditions.
Pathways
DNA polymerase kappa is integral to the translesion synthesis pathway and participates in the broader DNA damage response system. It relates functionally to other DNA polymerases including polymerase eta and zeta which together modulate cellular responses to DNA damage and repair. The interplay between these enzymes facilitates flexible and adaptive responses to various genotoxic insults highlighting their roles in maintaining genomic stability.
DNA polymerase kappa's association with carcinogenesis is of particular interest. Its error-prone activity can result in mutagenesis linking it to cancer development. POLK's expression or malfunction influences conditions like xeroderma pigmentosum where defective DNA repair increases cancer risk. Connections with the p53 pathway illustrate that defects in related DNA repair proteins can exacerbate the development of neoplastic diseases highlighting considerable implications for targeted cancer therapies.
