SMG6 also known as Suppressor with Morphological Effect on Genitalia 6 is an important component in the mRNA surveillance machinery and plays an important role in nonsense-mediated mRNA decay (NMD). It is a protein with a molecular mass of approximately 180 kDa. SMG6 exhibits endonuclease activity performing its function by cleaving mRNA substrates that contain premature stop codons. The protein is ubiquitously expressed across various tissues indicating its fundamental role in maintaining cellular functions.
Biological function summary
SMG6 associates with the core NMD factor UPF1 forming part of a complex involved in degrading faulty mRNA transcripts. This degradation process ensures that aberrant mRNAs do not translate into dysfunctional proteins essential for maintaining proteome integrity. SMG6 through its endonuclease activity precisely cleaves the target mRNA facilitating its decay and recycling safeguarding cellular homeostasis.
Pathways
SMG6 functions primarily within the nonsense-mediated decay (NMD) pathway a critical cellular quality control mechanism. This pathway intersects with other cellular processes such as mRNA metabolism and translation regulation. The protein interacts closely with both UPF1 and SMG7 which are essential proteins for efficient NMD. SMG6 ensures the NMD pathway operates effectively thereby maintaining cellular equilibrium against aberrant genetic events.
Aberrations in SMG6 function link to cancer and neurodegenerative disorders. Mutations or dysregulation of SMG6 may lead to an inability to eliminate faulty mRNAs contributing to the accumulation of defective proteins that play roles in tumorigenesis or neurodegeneration. In cancer altered interactions between SMG6 and proteins like UPF1 can result in unchecked cell proliferation. Similarly disruptions in SMG6-associated pathways might lead to neurodegenerative processes by allowing the accumulation of toxic protein products in neuronal tissues.
