MLK4 also known as Mixed Lineage Kinase 4 is a signaling protein involved in various cellular processes. It is a member of the MAP3K family and has a molecular weight of approximately 88 kDa. MLK4 is expressed in several tissues including the brain liver and kidney suggesting it plays roles in diverse physiological contexts. As a kinase it participates in phosphorylating specific substrates which impacts downstream signaling pathways.
Biological function summary
MLK4 influences cell proliferation differentiation and apoptosis through its kinase activity. It is part of signaling complexes that coordinate cellular responses to external stimuli linking surface receptors to the intracellular machinery. Through these roles MLK4 exerts important effects on cellular dynamics and maintenance of normal physiological balance.
Pathways
MLK4 integrates into the JNK and p38 MAPK signaling cascades which mediate responses to stress stimuli and regulate gene expression. MLK4 interacts with various other proteins including MLK3 to modulate these pathways effectively. Furthermore its role in these pathways positions MLK4 as an important player in ensuring cells react appropriately to potential damage or stress signals.
MLK4 has been associated with certain cancers and neurodegenerative diseases. Its involvement in cancer relates to its ability to influence cell growth and survival where MLK4 mutations or dysregulation might promote tumorigenesis. Moreover altered MLK4 signaling has links to Parkinson's disease particularly in conjunction with other kinases like LRRK2 suggesting its potential as a therapeutic target for modulating disease progression.
