PTIP also known as PAXIP1 is a protein that functions as a regulator in DNA repair and transcription. Its molecular mass is approximately 108 kDa. PTIP localizes in the nucleus and is expressed in various tissues with high expression in brain and testis. Mechanically PTIP participates in processes that maintain genomic stability and gene expression regulation. It interacts with other proteins to support its function in cellular DNA dynamics.
Biological function summary
PTIP plays critical roles in DNA damage response and chromatin remodeling. As a part of the MLL/COMPASS-like complex it facilitates the methylation of histone H3 on lysine 4 which is important for the transcriptional activation of genes. PTIP's action in DNA repair and transcription machinery emphasizes its involvement in the control of cell cycle progression and cellular homeostasis.
Pathways
PTIP integrates into significant processes including the homologous recombination and transcription activation pathways. It cooperates with proteins like ATM and 53BP1 in the DNA damage repair pathway ensuring efficient repair of double-strand breaks. By bridging these interactions PTIP contributes to the regulation of gene expression and maintenance of chromatin structure linking it to important cellular activities.
The dysfunction of PTIP associates with various cancer types due to its role in maintaining DNA integrity. Alterations in PTIP can lead to genomic instability which is a hallmark of tumorigenesis. Additionally PTIP's interaction with BRCA1 highlights its involvement in breast cancer where disruptions in DNA repair and cell cycle regulation are common pathological features.
