The Respiratory Syncytial Virus Subgroup A G Glycoprotein often called RSV G protein is a surface protein responsible for viral attachment to host cells. It weighs approximately 90 kDa and expresses on the surface of RSV Subgroup A. The G glycoprotein interacts with heparan sulfate proteoglycans on the surface of epithelial cells facilitating virus entry into host cells. Its structure includes two major regions: a central conserved domain and an N-terminal region which contribute to its function in attachment and pathogenesis.
Biological function summary
This glycoprotein plays an important role in the virus's ability to evade the host immune response. It is not part of a larger complex but acts independently to modulate immune system pathways. It does so by interfering with the host's innate immune signaling reducing the effectiveness of the host's initial defense mechanisms. This enables the virus to establish infection efficiently and replicate within the host resulting in respiratory illness.
Pathways
The RSV G glycoprotein is involved in the manipulation of host cell signaling pathways related to immune response and inflammation. The protein affects signaling through pathways such as NF-kB which is important for regulating immune responses. It also associates with proteins such as CX3CR1 a receptor involved in leukocyte migration and inflammatory response. This association helps RSV in evading immune surveillance and establishing infection within the host.
The RSV G protein is significantly linked to respiratory illnesses such as bronchiolitis and pneumonia. These conditions are especially severe in infants and elderly populations. The G protein's interaction with the CX3CR1 receptor involved in leukocyte trafficking contributes to the inflammation and immune system disruption found in these diseases. Understanding these interactions can aid in developing therapies targeting RSV infections reducing the burden of associated respiratory diseases.
