ZHX3 also known as Zinc Fingers and Homeoboxes 3 is a transcription factor with a molecular weight of approximately 100 kDa. It is composed of two C2H2-type zinc fingers and a homeobox domain which contribute to its DNA-binding abilities. ZHX3 expresses predominantly in the liver kidney and pancreas where it plays roles in regulating the expression of various genes. The protein's presence in these organs emphasizes its importance in controlling gene activity related to specific physiological functions.
Biological function summary
ZHX3 acts as a transcriptional repressor that works within multi-protein complexes to modulate gene expression. It interacts with other members of the ZHX family and operates in concert with a protein called NF-YA to affect the transcriptional landscape of a cell. Through these interactions ZHX3 impacts cellular processes such as differentiation growth and apoptosis. This protein's activity is essential for maintaining proper cellular functions and ensuring that cells respond correctly to internal and external cues.
Pathways
ZHX3 participates in vital transcriptional regulatory networks. These networks influence cell cycle control and apoptosis both important to normal cellular proliferation and overall homeostasis. In these pathways ZHX3 often interacts with proteins like ZHX2 and NF-YA forming complexes that facilitate its role as a transcriptional regulator. The function of ZHX3 is critical in these pathways to maintain the balance between cell survival and death.
Disruptions in ZHX3 function have implications in certain types of cancer notably hepatocellular carcinoma and renal cell carcinoma. Altered expression or mutation of ZHX3 can lead to dysregulated cell cycles and evasion of apoptosis contributing to tumorigenesis. In cancer ZHX3 interactions with proteins such as p53 and NF-YA become significant as they can influence tumor progression. Understanding ZHX3's role in these diseases can provide insights into potential therapeutic targets for drug development.
