Rapamycin is an antifungal metabolite produced by Streptomyces hygroscopicus . Rapamycin is a potent and specific mTOR inhibitor. This allosteric inhibitor of mammalian mTORC1 induces autophagy and also acts as an immunosuppressant. mTORC1 is a central integrator of nutrient signaling pathways and stimulates cell growth and proliferation by activating anabolic processes inhibiting catabolic process such as autophagy.

Formulation: 1 vial of Rapamycin (10mg, lyophilized), 1 vial of DMSO
Preparation: Solubility: DMSO: 18.28 mg/mL (20 mM) Preparation of stock solution: Resuspend 10 mg of lyophilized Rapamycin in 546.9 µL of fresh DMSO to make a 20 mM stock solution
Storage & Handling: Store lyophilized powder at -20°C. Protect from light. Once reconstituted, store aliquoted at -80°C. Use reconstituted material within 6 months to avoid loss of potency.

Recommended Usage: Rapamycin was used for induction of Autophagy in a range from 1 to 40 µM. It is recommended that the reagent be titrated for each experiment to determine optimal performance.
