Oligomycin A suppresses oxidative phosphorylation in mitochondria by inhibiting ATP biosynthesis. Oligomycin A binds to the proton channel on the F0 component of ATP synthase, decoupling the F0 and F1 factors of the ATP synthase complex and blocking the electron transport chain. This effect is useful for investigations including oxidative phosphorylation and other studies of cell metabolism.

Formulation: Lyophilized
Preparation: Solubility in DMSO: 100 mg/mL or 126.41 mM. Moisture-absorbing DMSO reduces solubility. Please use fresh DMSO. Resuspend 1 mg of lyophilized Oligomycin A in 250 µL DMSO or 100% ethanol to make a 5 mM stock solution. For the 500 µg size, resuspend in 125 µl DMSO or 100% ethanol to make a 5 mM stock solution.
Storage & Handling: Store lyophilized powder at -20°C, away from moisture. Once reconstituted, store aliquoted at -80°C. Use reconstituted material within 6 months to avoid loss of potency. Avoid repeated freeze-thaw cycles.
Application: Cell Culture - Verified
Recommended Usage: For inhibition of oxidative phosphorylation in primary immune cells (e.g. PBMCs or splenocytes) a range from 0.25 µM to 1.0 µM is recommended. It is recommended that the reagent be titrated for each experiment to determine optimal performance.
