Tau proteins constitute a family of six isoforms expressed by the MAPT (Microtubule-Associated Proteins/Tau) gene. Full-length natural tau protein consists of 441 amino acids (Tau-441, 2N4R). Tau isoforms are named based on the number of amino-terminal inserts (0N, 1N, and 2N) or carboxyl-terminal microtubule binding domain repeats (3R and 4R). Tau proteins are expressed mainly in the neurons of the central nervous system (CNS) in human brain. They help stabilize the internal skeleton of neurons in the brain. This internal skeleton has a tube-like shape through which nutrients and other essential substances travel to reach different parts of the neuron. Tau proteins are important biomarkers for neurodegenerative disease research such as Alzheimer’s disease (AD) and Parkinson disease (PD).
