Mouse perforin, encoded by the PRF1 gene, is a pore-forming protein found in the granules of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. When these immune cells encounter a target cell, such as a virus-infected or cancerous cell, they release perforin, which then forms pores in the target cell’s membrane. This allows other molecules, like granzymes, to enter the target cell and induce apoptosis , effectively killing the harmful cell. A mouse perforin molecule consists of three domains: a Membrane Attack Complex/Perforin (MACPF) domain at the N-terminus, a C2 domain which required Ca2+ ions responsible for membrane docking, and an Epidermal Growth Factor (EGF)-like domain; these domains are crucial for its pore-forming function on target cell membranes. Perforin plays a crucial role in the immune system’s ability to eliminate infected or malignant cells and maintain overall immune regulation. Deficiencies in perforin can lead to severe immune disorders and increase susceptibility to infections and cancers.
