S100A10 is a member of the S100 family of proteins found in vertebrates with over 20 known members. S100A10 primarily functions as a regulator of membrane-related processes, including endocytosis and exocytosis, by forming a complex with annexin A2. This complex facilitates the trafficking and surface expression of ion channels, such as the epithelial sodium channel and certain TRP channels. S100A10 is also involved in plasminogen activation and fibrinolysis by promoting the generation of plasmin on the cell surface. Additionally, it has roles in modulating serotonin signaling and has been implicated in mood regulation and depression.

Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 0.0625 - 2.5 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 2.5 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.015 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following: -Fixation Buffer (Cat. No. 420801) with 100% ice-cold methanol -100% ice-cold methanol only Fixation/permeabilization with Fixation Buffer followed by 0.5 % Triton X-100 is not recommended due to dim signal. For use in intracellular flow cytometry (ICFC), we recommend fixation/permeabilization using fixation buffer (Cat. No. 420801) followed by True-Phos™ Perm Buffer (Cat. No. 425401). Fixation/permeabilization with either Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-NuclearTM Transcription Factor Buffer Set (Cat. No. 424401) is not recommended.
