Biolegend, 649451, Purified anti-BBC3/PUMA Antibody, 25μg

BBC3, also known as p53 upregulated modulator of apoptosis (PUMA), is a member of the Bcl-2 family. BBC3 promotes apoptosis and can be transcriptionally regulated by p53. When p53 is activated by survival signals, the increased levels of BBC3 induce apoptosis through mitchondrian dysfunction where BBC3 proteins are localized. BBC3 can also induce apoptosis in a p53-independent manner due to other stimuli such as cell stress. p53-mediated apoptosis and tumor suppression are impaired by targeted disruption of the BBC3 gene. There are four different isoforms of BBC3 (α, β, γ, and δ) but only the α and β forms interact with Bcl-2 and Bcl-xL, localize to the mitochondria, and promote apoptosis.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This antibody (clone W22198A) does not cross-react with mouse BBC3. This product is not suggested for use in mouse.
Additional Product Notes: For immunocytochemistry (ICC), we recommend to use 4% PFA fixation followed by permeabilization with ice cold-methanol, or ice-cold methanol fixation. This antibody (clone W22198A) is not suitable for IHC.
