Product Description

TNFR-associated factor 3, also known as TRAF3, is a member of the TNF receptor associated factor (TRAF) protein family and was originally identified as an adaptor molecule binding to the cytoplasmic tail of CD40. The engagement of CD40 with its ligand recruits the complex, which consists of TRAF3, TRAF2, and c-IAP1/2. This results in the stabilization of NF-κB inducing kinase (NIK). NIK phosphorylates the IKKα complex and subsequently leads to the activation of the RelB/p52 complex. TRAF3 is involved in the Toll-like receptor (TLR) mediated activation of proinflammatory genes such as type I interferons and IL-10. Pathogens detected by TLR3 and TLR4 recruit the adaptor protein TRIF. In turn, this activates IRF3 via the TRAF3-TBK1-IKKe complex. TRAF3 has been shown to play important roles in the regulation of innate immune response, antibody class switching, and B cell survival.
