Product Description

CD344 is a member of the Frizzled family, also known as Frizzled-4 (FZD-4). Ten Frizzled family members have been identified in humans. According to the identity of amino acid sequences, frizzled proteins can be divided into four subgroups: subfamily 1 (FZD-1, FZD-2, and FZD-7), subfamily 2 (FZD-5 and FZD-8), subfamily 3 (FZD-9 and FZD-10), subfamily 4 (FZD-4) and subfamily 5 (FZD-3 and FZD-6). The human FZD-4 gene is located on chromosome 11q14–q21. FZD-4 is a 48–53 kDa seven-transmembrane G protein–coupled receptor (GPCR) that serves as a receptor for Wnt proteins and Norrin. In the presence of Wnt ligand, Wnt binds to extracellular cysteine rich domain (CRD) of Frizzled. A receptor complex forms between Frizzled and low-density lipoprotein receptor-related protein 5/6 (LRP5/6). Norrin, an atypical Wnt ligand, binds to FZD-4 and LRP5/6, activating the canonical Wnt/β-catenin pathway (canonical Wnt signaling). FZD-4 is a neuronal stem cell marker and is also expressed in fetal neuronal progenitor cells and intestinal neuronal cells. High levels of FZD-4 expression are found in the kidney, lung, brain, and liver. FZD-4 has been reported to regulate retinal and inner ear angiogenesis and plays a role in embryonic development, tissue and cell polarity, and cell proliferation.

Source: Recombinant human Frizzled-4, amino acid Phe37-Glu180 (Accession No. Q9ULV1), with a proline at N-terminus and a C-terminal Xa-mutant hFc (C103S), was expressed in CHO cells.
Molecular Mass: The 382 amino acid recombinant protein has a predicted molecular mass of approximately 43.0 kD. The DTT-reduced and non-reduced proteins migrate at approximately 50 - 62 kD and 80 - 115 kD by SDS-PAGE respectively. The N-terminal amino acid is Pro.
Purity: > 95%, as determined by Coomassie stained SDS-PAGE.
Formulation: 0.22 µm filtered protein solution is in PBS, pH 7.2
Endotoxin Level: Less than 0.1 EU per µg protein as determined by the LAL method.
Concentration: 50 µg size is bottled at 0.2 mg/mL
Storage & Handling: Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity: ED50= 0.25 - 2.5 µg/mL as measured by its ability to inhibit Topflash reporter activity in HEK293T human embryonic kidney cells induced by recombinant human Wnt3a.

Q: What is the specific activity or ED50 of my recombinant protein?
A: The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
Q: Have your recombinants been tested for stability?
A: Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
