Replication protein A (RPA) is a single-stranded DNA-binding protein comprised of three subunits of 70, 32, and 14 kD, and is involved in the regulation of DNA replication, repair, and homologous recombination. During replication, RPA stabilizes single-stranded DNA by protecting it from nucleases and preventing the formation of secondary structures. Under genotoxic stress conditions, the RPA32 subunit is phosphorylated by the PI3K family of kinases including ATM, ATR, and DNA-PK. Hyperphosphorylated RPA32 has shown to promote homologous recombination by recruiting Rad51, which is required for homologous pairing and strand exchange reactions during DNA repair.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This clone does not recognize mouse RPA2.Triton X-100 permeabilization produced stronger staining than methanol permeabilization in ICC testing.
