HIGD1A and HIGD2A (of 10.4 and 11.5 kDa, respectively) have two hydrophilic transmembrane domains and are localized in the inner mitochondrial membrane. HIGD1A and HIGD2A expression is induced under stress conditions like hypoxia or glucose deprivation in human cervical epithelial cells and murine cerebral cortical neurons. HIGD1A may play pleiotropic roles in mitochondrial respiratory chain biogenesis. HIGD2A forms a 50 kDa regulatory module with COX4-1, COX5B, and COX6A1, which binds to newly synthesized COX3 to promote its binding to the previously assembled COX1 + COX2 module (PMID:33291261,32375044).
