It is initially expressed as a large polyprotein precursor, which is subsequently proteolytically processed into individual functional filaggrin molecules. The free filaggrin binds to keratin intermediate filaments, causing their aggregation into macrofibrils in which the intermediate filaments are aligned in tightly packed parallel arrays.
