EHMTs, along with LSD1 (an H3K4 demethylase), are components of the REST complex, which silences neuronal pathway genes in non-neuronal cells. EHMT1 is mainly responsible for the monomethylation and dimethylation of histone H3 lysine 9 (H3K9), forming a heteromeric complex with EHMT2 (G9a) in euchromatin (PMID: 34214254, 37663929).
