Human SAA ELISA Kit (SAA1) ab100635 is a sandwich ELISA to measure Human SAA (SAA1) in serum, plasma, cell culture supernatant with a sensitivity of 0.5 ng/ml.
How the assay works
This assay employs an antibody specific for Human Serum Amyloid A coated on a 96-well plate. Standards and samples are pipetted into the wells and Serum Amyloid A present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Serum Amyloid A antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Serum Amyloid A bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

1. Add standard or sample to each well used. Incubate at room temperature
2. Wash and add prepared biotinylated antibody to each well. Incubate at room temperature.
3. Wash and add prepared streptavidin solution. Incubate at room temperature.
4. Add TMB Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.
Optimization may be required with urine samples. ab100635 cross reacts with SAA2.

Serum Amyloid A 1 (SAA1) sometimes called acute phase serum amyloid is a 12-kilodalton protein. It is mainly produced by the liver in response to pro-inflammatory cytokines during the acute-phase response. SAA1 is highly expressed in the liver and released into the bloodstream under inflammatory conditions. Other tissues such as adipocytes and epithelial cells can also express SAA1 to a lesser extent. This protein plays a role in lipid metabolism and immune system regulation.
Biological function summary
Serum Amyloid A 1 acts as an apolipoprotein associated with high-density lipoprotein (HDL) in plasma. It contributes to cholesterol metabolism by removing cholesterol from tissues and assisting in its transport to the liver for excretion. SAA1 also plays a role in modulating immune cell function including recruiting immune cells to sites of inflammation and inducing enzymes that degrade the extracellular matrix. It is not a part of a larger complex but interacts dynamically with lipid and immune system elements to exert its effects.
Pathways
Its function is integrated into both inflammation and cholesterol metabolism pathways. During inflammation SAA1 modulates the activity of enzymes such as secretory phospholipase A2 to enhance the breakdown of phospholipids. In the cholesterol metabolism pathway SAA1 interacts closely with proteins involved in reverse cholesterol transport. Apolipoprotein A-I is an associated protein that functions alongside SAA1 in remodeling HDL particles and mediating cholesterol efflux from tissues.
Serum Amyloid A 1 has associations with chronic inflammatory diseases and amyloidosis. Persistent elevated levels of SAA1 can contribute to the development of amyloid deposits in tissues a feature of amyloidosis. Rheumatoid arthritis is another condition where SAA1 levels significantly rise linking it to joint inflammation and damage. The protein connects with interleukin-6 a cytokine that drives the inflammatory response found in these diseases.
