The protein P4HA3 also known as Prolyl 4-Hydroxylase Subunit Alpha 3 functions as an enzyme involved in collagen synthesis. It is part of the prolyl 4-hydroxylase complex which catalyzes the hydroxylation of proline residues in collagen aiding its stability and structural integrity. P4HA3 has a molecular mass of approximately 61 kDa. Expression of this protein occurs widely especially in connective tissues where collagen is a major component.
Biological function summary
P4HA3 plays an important role in remodeling and maintaining the extracellular matrix through its contribution to collagen biosynthesis. As a member of the prolyl 4-hydroxylase complex it partners with beta subunits to form a functional enzyme. This association is critical for hydroxyproline formation which is important for assembling stable collagen triple helices. The enzyme's activity impacts cellular processes such as migration adhesion and growth embedded in tissue repair and development.
Pathways
P4HA3 is an integral part of the collagen biosynthesis pathway. This pathway includes the post-translational modification of collagen proteins which is essential for producing functional collagen fibrils. It works in concert with other proteins like P4HA1 and P4HB which are also involved in collagen hydroxylation. The activity of P4HA3 impacts the TGF-beta signaling pathway which influences various cellular functions including proliferation and differentiation.
Alterations in P4HA3 are linked to fibrosis and cancer. Fibrosis involves excessive collagen deposition often driven by increased activity of P4HA3 leading to tissue scarring and functional impairment as observed in liver and lung conditions. In cancer upregulation of P4HA3 can promote tumor proliferation and metastasis changing the tumor microenvironment to favor cancer cell survival. P4HA3’s interaction with P4HB can enhance these pathological processes highlighting its potential as a therapeutic target.
