Abcam's Flt3 Ligand Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of mouse Flt3 Ligand in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for mouse Flt3 Ligand coated on a 96-well plate. Standards and samples are pipetted into the wells and Flt3 Ligand present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse Flt3 Ligand antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Flt3 Ligand bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Flt3 Ligand also known as FLT3LG or FL cytokine is a protein primarily involved in hematopoiesis which is the process of forming blood cellular components. It has a molecular weight of approximately 23 kDa. Flt3 Ligand binds to Flt3 receptor tyrosine kinase leading to the proliferation of hematopoietic progenitor cells. It is mainly expressed in bone marrow stromal cells peripheral tissues and thymus. The interaction between Flt3 Ligand and its receptor triggers a cascade of intracellular signaling promoting various cellular outcomes essential for maintaining immune and hematopoietic homeostasis.
Biological function summary
The Flt3 Ligand is a critical stimulator for growth and differentiation of stem cells and progenitors in the immune system. Although not part of a large multi-protein complex it directly regulates the early stages of dendritic cell and B cell development by enhancing their proliferation and differentiation. Additionally Flt3 Ligand's involvement extends to the activation of progenitor cells responsible for natural killer cells contributing significantly to innate immunity. Its action ensures the production and survival of cells necessary for an efficient immune response.
Pathways
Flt3 Ligand plays a significant role in the hematopoietic and immune signaling cascades. It interacts through the Flt3 receptor pathway which parallels with pathways mediated by the c-Kit and GM-CSF receptors. These pathways involve signal transduction mechanisms that promote survival proliferation and differentiation of hematopoietic progenitor cells. In particular the PI3K/AKT and RAS/RAF/MEK pathways are activated downstream of Flt3 Ligand binding leading to cell cycle regulation and anti-apoptotic functions. Its integration with multiple signaling pathways ensures coordinated development and function of immune cells.
Flt3 Ligand's dysregulation links it to acute myeloid leukemia (AML) and immune disorders such as aplastic anemia. Abnormal expression and activity of Flt3 Ligand contribute to the proliferation of leukemic cells in AML by interacting with mutated forms of Flt3 receptor. Through this relationship Flt3 Ligand fosters the growth of malignant hematopoietic cells exacerbating the disease. Similarly in aplastic anemia altered Flt3 Ligand expression impairs normal hematopoiesis leading to ineffective immune cell production. Understanding these interactions positions Flt3 Ligand as a potential target for therapeutic interventions in these debilitating conditions.
