Mkl1 also known as MRTFA is a transcriptional coactivator that plays a role in regulating gene expression. It has a molecular weight of approximately 142 kDa. Mkl1 is widely expressed in various tissues including the heart lung and skeletal muscle. As a coactivator it interacts with serum response factor (SRF) to facilitate the transcription of target genes involved in cytoskeletal dynamics and cell differentiation processes.
Biological function summary
Mkl1 is important in the regulation of actin cytoskeleton remodeling. It does not act alone; it forms part of a dynamic complex with actin monomers. These interactions are essential for the modulation of gene transcription. Mkl1 binds to gene promoters enabling transcriptional activation in response to cellular signals that involve changes in actin polymerization.
Pathways
Mkl1 integrates into the RhoA signaling pathway and the Hippo pathway important for cytoskeleton organization and cell growth control respectively. Within these pathways Mkl1 regulates the activity of SRF coordinating cellular responses to mechanical stress and environmental stimuli. Mkl1 also interacts with other proteins such as myocardin and its family members which are relevant in smooth muscle cell differentiation.
Mutated or dysregulated Mkl1 has been connected to certain leukemias particularly acute megakaryoblastic leukemia. It also relates to cardiac hypertrophy a condition associated with abnormal heart muscle growth. Mkl1 dysregulation leads to altered SRF pathway activity impacting the expression of genes involved in cell growth and differentiation which is critical in these disease processes.
