ESCO2 also known as Establishment of Sister Chromatid Cohesion N-Acetyltransferase 2 is a protein with a mass of about 68 kDa. It is expressed widely across various human tissues with notable presence in proliferating cells. ESCO2 has a mechanistic role in the establishment of sister chromatid cohesion during the S phase of the cell cycle. It works by acetylating the cohesin subunit SMC3 which is essential for proper chromosome segregation during mitosis and meiosis.
Biological function summary
ESCO2 functions as a part of the chromosomal cohesion complex which is necessary for maintaining genome stability. The cohesion complex ensures that sister chromatids remain paired from DNA replication in S phase until anaphase. This complex requires synchronized action of cohesin proteins including STAG2 and RAD21 which work together with ESCO2 to ensure the fidelity of chromosomal transmission. By facilitating proper cohesion ESCO2 prevents aneuploidy which is often incompatible with cell viability.

The protein ESCO2 interacts with the cell cycle regulatory pathways and chromosomal dynamics. Within these pathways ESCO2 cooperates with proteins like CDK1 and PLK1.
