ID4 also known as Inhibitor of DNA Binding 4 is a member of the ID protein family that inhibits basic helix-loop-helix (bHLH) transcription factors. The protein ID4 has a mass of approximately 16 kDa. It lacks a DNA binding domain but can still regulate transcription by forming heterodimers with other bHLH proteins preventing their interaction with DNA. ID4 is expressed in various tissues with notable expression in the central nervous system mammary gland and testis.
Biological function summary
ID4 plays a significant role in regulating cell proliferation differentiation and apoptosis. It acts within the context of a protein complex by inhibiting the activity of bHLH transcription factors involved in these processes. It maintains stem cell properties by preventing premature differentiation especially in neural progenitor cells. ID4 also participates in the development of several tissues including regulating the differentiation of myoblasts during muscle development.
Pathways
ID4 is integral to the cell cycle and development pathways. It interacts with the p53 pathway by modulating transcription factors involved in cell cycle control and apoptosis. Additionally ID4 has connections with the Notch signaling pathway which is important for cell fate determination and differentiation. In these pathways ID4 often associates with HES and HEY proteins which are downstream transcriptional regulators influenced by Notch signaling.
Aberrant expression of ID4 is linked to several cancers including glioblastoma and breast cancer. In these diseases ID4 can either promote or inhibit tumor growth depending on the context and its interaction with other proteins like p21 which is a cyclin-dependent kinase inhibitor involved in cell cycle regulation. Furthermore changes in ID4 expression are implicated in neurodegenerative disorders due to its regulation of neural stem cell differentiation and survival.
