The P2X2 receptor also called P2RX2 is a ligand-gated ion channel that opens in response to the binding of extracellular ATP. It belongs to the P2X receptor family and has a molecular weight of approximately 51 kDa. P2X2 is expressed in a variety of tissues most notably in the nervous system including the brain and spinal cord as well as other excitable tissues like skeletal muscle. Its presence contributes to the regulation of synaptic transmission and neuromuscular communication.
Biological function summary
P2X2 participates in the modulation of cellular excitability and neurotransmitter release. It forms homomeric channels or heteromeric channels by assembling with other P2X subunits such as P2X3. Its role becomes significant in the context of the central and peripheral nervous systems where it influences ion flow particularly for sodium potassium and calcium across the cell membrane. The ion flow alters the membrane potential and affects neuronal firing and signal propagation.
Pathways
P2X2 integrates into the purinergic signaling pathway and plays a part in synaptic transmission modulation. It influences various biological processes through interactions with related proteins such as other members of the P2X family like P2X4 and P2X7. These interactions fine-tune cellular responses to ATP in the nervous system and are important for maintaining physiological processes like neurotransmission and pain perception.
P2X2 associates with conditions like neuropathic pain and hearing loss. The receptor's function connects to auditory pathways where its dysfunction might contribute to cochlear disorders. Additionally through its role in purinergic signaling P2X2 links with other proteins involved in pain pathways such as P2X3 whose modulation could provide therapeutic targets for pain management.
