ADCK3 also known as AarF domain-containing kinase 3 or COQ8A is a protein kinase with an approximate mass of 68 kDa. It is largely expressed in the mitochondria which indicates its role in cellular energy processes. The human ADCK3 gene encodes this protein and it serves a critical function in the biosynthesis of coenzyme Q10 an essential component of the mitochondrial electron transport chain. ADCK3's structure includes a kinase domain hinting at its enzymatic activity in phosphorylation processes.
Biological function summary
ADCK3 participates in the regulation of coenzyme Q10 levels an essential lipid-soluble antioxidant. The protein is a part of the mitochondrial electron transport chain complex contributing to the production of ATP. As a kinase ADCK3 influences the modulation of the complex's components through phosphorylation stabilizing the function and efficiency of energy conversion processes within mitochondria. Its role is critical for maintaining cellular energy balance and reducing oxidative stress.
Pathways
ADCK3 engages in the coenzyme Q10 biosynthesis pathway. It closely interacts with COQ proteins like COQ9 orchestrating the synthesis and function of the electron transport chain. Additionally it takes part in mitochondrial kinase pathways that ensure proper mitochondrial function. The cooperation between ADCK3 and other proteins such as COQ6 emphasizes its importance in maintaining mitochondrial integrity and energy metabolism.
Mutations in the ADCK3 gene have been linked to Coenzyme Q10 deficiency and spinocerebellar ataxia. Individuals with mutations often face progressive cerebellar atrophy a condition that affects motor skills and movement coordination. The association of ADCK3 with COQ6 in these disorders highlights its influence in the pathological development and underlines the potential therapeutic interest in modulating its activity.
