NCM also known as Neuronal Calcium Sensor-1 (NCS-1) is a protein involved in calcium-binding. It has a molecular mass of approximately 22 kDa and features three EF-hand motifs essential for its function. NCM mainly expresses in the central nervous system including areas like the cerebellum and hippocampus. It also finds expression in peripheral tissues such as the heart and adrenal gland indicating its relevance in different physiological processes.
Biological function summary
NCM modulates intracellular calcium signaling influencing processes like neurotransmitter release and synaptic plasticity. It may also participate in a complex with other proteins that affect calcium-dependent processes. NCM's role includes the regulation of ion channels and membrane trafficking events important for sustained neuronal activity. It impacts the communication between neurons and the overall nervous system function.
Pathways
NCM acts within the calcium signaling pathway and is linked to the phosphoinositide signaling pathway. NCM interacts with proteins such as the Dopamine Receptor D2 and PI4KIIIβ which are important in these pathways. This interaction allows NCM to influence various cellular responses initiated by changes in intracellular calcium levels governing diverse signaling outcomes.
NCM has associations with neurological conditions such as schizophrenia and bipolar disorder. Altered NCM function may affect calcium homeostasis in neurons contributing to these disorders. In the context of schizophrenia NCM interacts with the Dopamine Receptor D2 altering dopamine signaling implicated in the disease. Understanding NCM's relationship to these conditions can reveal potential therapeutic targets for intervention.
